The orchestra consists of 21 or 22 members and is named after the Church of St. Luke in Manhattan, where the ensemble first performed. In 1984 Carnegie Hall invited the Orchestra of St. Luke’s to perform as part of their Handel Opera Festival. With the orchestra's success that season, the Orchestra of St. Luke’s was invited to return each year in an annual subscription series.

The Orchestra of St. Luke’s strives to bring music to everyone interested and prioritizes accessible education programs for their community. They provide a space for the community to rehearse, perform, and record through the DiMenna Center for Classical Music.
